Cover Illustration: Fluorescence 3D reconstruction of the Nkx2-5+/GFP mouse embryonic heart. GFP was targeted into the Nkx2-5 locus resulting in GFP expression specifically in cardiomyocytes (Biben C, Weber R, Kesteven S, Stanley E, McDonald L, Elliott DA, Barnett L, Köentgen F, Robb L, Feneley M, et al. 2000. Cardian septal and valvular dysmorphogenesis in mice heterozygous for mutations in the homeobox gene Nkx2-5. Circ Res 87: 888-895). Embryos were harvested at E9.5; heart tissue was dissected and fixed in 4% paraformaldehyde. A z-stack was acquired with a Zeiss LSM510 META confocal microscope using a 20X objective and a reconstructed 3D projection was generated using the LSM software.
