Dewaxing and Rehydrating Sections prior to In Situ Hybridization

INTRODUCTION

This protocol describes how to remove wax from embryo or tissue sections that have been affixed to glass slides. Traditionally, xylene has been used for this purpose, but less toxic solutions can also be employed. The embryo or tissue sections are then progressively rehydrated for compatibility with subsequent alcohol stains, aqueous stains, immunohistochemistry, or in situ hybridization.
