Collecting Two-Cell- to Compacted Morula-Stage Embryos

INTRODUCTION

This protocol describes a method for collecting two-cell- to compacted morula-stage embryos. Such embryos can then be used for microinjection.


MATERIALS

Reagents

Ethanol, 70%

recipe M2 medium at room temperature

Pregnant female mice (20-60 hours post coitum) sacrificed humanely (See Quick and Humane Sacrifice of a Mouse by Cervical Dislocation)

Equipment

Embryo-handling pipette consisting of mouth or hand-held pipette assembly and pulled capillary (see Making Pipettes from Hard Glass Capillaries and Preparing Siliconized Pipettes)
