INTRODUCTION
This protocol describes the preparation of genomic DNA from a 5-ml liquid culture of yeast cells.
MATERIALS
Reagents
Ammonium acetate, 5M
Appropriate yeast cultures
Chloroform:isoamyl alcohol(24:1, v:v)
EDTA-Sark
Ethanol, 95%, cold
Ethanol, 70%
Growth medium, rich or selective (see Step 1)
Yeast lysis buffer II
NaCl, 5 M
Phenol , equilibrated with H2O
