Miller LB agar plates

MATERIALS

Reagents

Antibiotic of choice

Ampicillin sodium sulfate (50 mg/mL; Sigma-Aldrich)

Kanamycin monosulfate (30 mg/mL; Fisher Scientific)

Filter-sterilize and store antibiotics at -20°C.

LB agar, Miller (Sigma-Aldrich)

Equipment

Aluminum foil

Autoclave

Flask, 2-L

Petri dishes, 100- × 10-mm

Plastic bags

Tape

Thermometer

METHOD

  • 1. Dissolve 40 g of Miller LB agar in 1 L of H2O in a 2-L flask. Cover with foil and autoclave.

  • 2. Monitor the temperature as the solution cools. When the temperature reaches ~ 50°C, add 1 mL of the kanamycin or ampicillin stock solution.

  • 3. Pour ~10 mL into each Petri dish. Swirl to coat the plates. Allow the agar to solidify at room temperature.

  • 4. Place plates into a plastic bag. Seal with tape. Store at 4°C.
