Rich auto-induction medium

β-lactose (Sigma-Aldrich) 10 g 1%
Glucose (Sigma-Aldrich) 0.5 g 0.05%
Glycerol (100%; Fisher Scientific)a 5 g 0.5%
KH2PO4 (Fisher Scientific) 6.8 g 50 mM
MgSO4 (1 M; Fisher Scientific)b 2 mL 2 mM
N-Z amine AS (Sigma-Aldrich) 10 g 1%
Na2HPO4 (Fisher Scientific) 7.1 g 50 mM
Na2SO4 (Fisher Scientific) 0.71 g 5 mM
NH4Cl (Fisher Scientific) 2.67 g 50 mM
Trace metals (1000X; Sigma-Aldrich)a 400 μL 0.2X
Yeast extract (Fisher Scientific) 5 g 0.5%
Adjust the pH to 8.0-8.2.

This medium is based on Studier’s medium ZYM-5052, with modifications for high-level protein production (Studier FW. 2005. Protein production by auto-induction in high density shaking cultures. Protein Expr Purif 41: 207-234).
