Use a standard YPD recipe supplemented with a 200 mg/liter effective concentra¬tion of G418 disulfate salt (Sigma). The G418 powder can be added direct¬ly to the plate medium before pouring. Note that because of phenotypic lag, you must allow the cells to recover from the transformation for 4 hours- overnight before plating.
