Hydroxyapatite column packing buffer

A 200 mM sodium phosphate dibasic solution (pH 9-10) is recommended for packing CHT ceramic hydroxyapatite. Packing columns or preparing a slurry from dry CHT with solutions containing <50 mM sodium phosphate can cause cloudiness in the supernatant and should be avoided. Solutions of >200 mM sodium phosphate may be used to slurry dry CHT and pack columns but offer no additional benefit.

Do not use anhydrous sodium phosphate because these salts contain levels of pyrophosphate that prevent binding of some macromolecules. Monobasic sodium phosphate monohydrate and dibasic sodium phosphate 7H2O are recommended.
