Agarose sealing solution

20 ml 5X SDS electrophoresis buffer

0.5 g agarose

Use a low electroendosmosis form of agarose. A low-melting-temperature form may also be advisable.

A few grains of bromophenol blue

80-ml H2O

Add all ingredients to a 500-ml Erlenmeyer flask and swirl to suspend. Heat in a microwave oven on low setting until the agarose is completely dissolved. It is best to do this in 20-second intervals to ensure that the solution does not boil over. Dispense in 1.5-ml aliquots in screw-cap tubes and store at room temperature or at 4ºC.
