Mowiol mounting medium for FRET analysis

Glycerol

Mowiol 4-88 (Calbiochem)

Tris-Cl (200 mM, pH 8.5)

Mix 6 ml of glycerol, 2.4 g of Mowiol 4-88, and 6 ml of distilled, deionized H₂O. Initially shake for 2 hours at room temperature, then add 12 ml of 200 mM Tris-Cl (pH 8.5) and incubate at 50°C with occasional mixing until the Mowiol dissolves (~3 hr). Filter through a 0.45-μm membrane filter and store in aliquots at 4°C for weeks, or -20°C for months.
