Root-inducing medium (RIM)

1X MS (Murashige and Skoog) salts

10 g sucrose

0.5 g 2-[N-morpholino]ethanesulfonic acid (MES)

KOH (1 N)

Agarose

1 ml Gamborg’s B5 vitamins (1000X)

1 ml α-naphthylphthalamic acid (NAA) (1000X)

500 mg timentin (optional, see below)

500 mg carbenicillin may be used in place of timentin

For 1 liter RIM, add MS salts, sucrose, and MES. Adjust pH to 5.7 with KOH, and add agarose to 0.5% (w/v). After autoclaving, cool medium to ~60ºC and then add Gamborg’s B5 vitamins and NAA (final concentration 2 mg/liter). If there are problems with Agrobacterium growth, use RIM with 500 mg/liter timentin or carbenicillin.
