FIONA imaging buffer with Trolox

Reagent Quantity
10% glucose 4 μL
1 M MgCl2 1 μL
Trolox solution (saturated) 94 μL
Gloxy 1 μL
Add the Gloxy just prior to using the imaging buffer. The final volume is 100 μL.
At high concentrations, Trolox can interfere with motility; therefore, 40% Trolox is recommended for motility experiments. To prepare 40% Trolox, decrease Trolox in the recipe above to 40 μL, and add:
54 μL of T50 buffer, or
54 μL of M5 buffer (for myosin assays).
