Recipe

Reagent	Quantity
1-2.5 mg/mL CellTracker CM-DiI (Molecular Probes), prepared in DMSO and stored at −20°C.	5 μL
0.3 M sucrose, filter-sterilized (not autoclaved)	25 μL
Protect the working solution from light by wrapping the container with aluminum foil and keep it at 4°C.
Before using, test the dye solution by putting a drop on a glass coverslip under light paraffin oil. The dye should stay in a drop form and not spread under the light oil. If the dye seeps into the light paraffin oil, prepare a fresh aliquot from the stock solution.
