Removing the Vitelline Membrane from Xenopus laevis Embryos

This protocol was adapted from “Preparing Embryos for Manipulation,” Chapter 6, in Early Development of Xenopus laevis by Hazel L. Sive, Robert M. Grainger, and Richard M. Harland. Cold Spring Harbor Laboratory Press, Cold Spring Harbor, NY, USA, 2000.

INTRODUCTION

This protocol presents a method for the removal of the vitelline membrane from Xenopus embryos, which is essential for embryo dissection experiments. Removal of the vitelline membrane is a technique that becomes easier with practice. The easiest embryos on which to learn this technique are those with a large space between the membrane and the embryo, that is, late neurula stages.
