Sater’s modified blastocoel buffer (SMBB)

Reagent Amount to add
5 M NaCl 9.90 ml
Gluconic acid, sodium salt 7.95 g
NaHCO3 0.53 g
1 M KCl 4.5 ml
2.5 M CaCl2 0.4 ml
1 M MgSO4 1 ml
6 mM HEPES to pH 8.1
Gentamycin sulfate to 50 μg/ml
H2O to 1 liter
If desired, standard amounts of penicillin and streptomycin and 1 mg/ml BSA may also be added. Filter-sterilize the solution. Store it frozen in 50-ml aliquots.
