Agarose (1.5%) for neocortex imaging
0.375 g agarose (low melting point; Sigma)
Warm 0.375 g of low-melting-point agarose in 25 mL of sterile cortex buffer until fully dissolved and clear of precipitate. Prepare this solution fresh and sterile for each experiment and keep it hot with mixing during the procedure. Before applying the hot agarose to the cortex, allow it to cool briefly, and check that the temperature is not above ~37°C, by squirting a drop onto the back of the hand.
