METHOD

1. Working under a fume hood, in a glass scintillation vial, mix 0.92 g of paraformaldehyde in 2.5 mL of H2O and 35 μL of 1N KOH.

2. Dissolve by heating on a hot plate while stirring with a magnetic stir bar.

3. Cool the solution on ice.

4. Filter through a 0.45-μm syringe filter.

5. It is best to prepare the solution fresh on the day of tissue fixation, although it may be stored at 4°C for up to 1 wk. The recipe can be scaled up, if necessary.
