Construction of the random DNA library. Oligonucleotides with 50 random nucleotide positions ([N]50), a 5′-flanking region containing an AscI restriction site (blue), and a palindromic 3′-flanking region containing a BamHI restriction site (red), are self-annealed, rendered double-stranded, and cloned into the AscI site of a bacterial plasmid vector.
