Hematal-16

MATERIALS

Reagents

Aluminum sulfate (Al2[SO4]3•18H2O)

Prepare an aqueous solution by dissolving 8 g aluminum sulfate in H2O to a final volume of 500 mL.

Ethylene glycol

Hematoxylin

METHOD

  • 1. Dissolve 0.5 g of hematoxylin in a mixture of H2O (125 mL) and ethylene glycol (125 mL). Bubble air through the solution for ~4 wk. Every few days, make up again to 250 mL with H2O to compensate for evaporation.

  • 2. Add to the above solution 250 mL of ethylene glycol, followed by 500 mL of an aqueous solution of aluminum sulfate. The final volume is 1000 mL.

    When the hematein is made by atmospheric oxidation, as described here, the hematal-16 solution is stable for ~2 yr.
