Reagents

Blocking reagent (Roche Applied Science, 11096176001)

Sheep serum (heat-inactivated)

To prepare, incubate stocks of sheep serum for 1 h at 56°C. Aliquot into smaller tubes and store at −20°C.

METHOD

  • 1. Add 2 g of blocking reagent to 100 mL of 1X MBST. Heat and agitate on a magnetic stirrer for 1 h to allow the blocking reagent to go into solution.

  • 2. To prepare 50 mL of blocking buffer, combine 10 mL of heat-inactivated sheep serum with 40 mL of the blocking agent solution from Step 1.
