Ficoll cushion solution

Ficoll 400 (Sigma F8016)

PE buffer (50X)

Triton X-100 (Sigma-Aldrich T9284), 10% (w/v) in H₂O

• 1. Prepare 30 mL of 1X PE buffer containing 0.1% (w/v) Triton X-100.

• 2. Add the PE/Triton solution slowly to 5 g of Ficoll until the total mass is 25 g.

• 3. Stir for several hours at room temperature.

• 4. Store at 4°C.
