Cresyl violet acetate stain

MATERIALS

Reagents

Cresyl violet, 1% (w/v) in H2O

Glacial acetic acid

Sodium acetate

Equipment

Aspirator pump for filtering

Whatman #1 filter paper

METHOD

  • 1. Add 6 mL of glacial acetic acid to 994 mL H2O to make Solution A.

  • 2. Add 13.6 g of sodium acetate to 1000 mL H2O to make Solution B.

  • 3. Combine Solutions A and B in a 9:1 ratio and titrate to pH 3.7 to make an acetate buffer.

  • 4. Combine the acetate buffer with 1% cresyl violet in a 1:1 ratio to make cresyl violet acetate stain.

  • 5. Filter the staining solution with an aspirator pump and Whatman #1 filter paper.
