Recipe

Shoot-inducing medium (pH 5.8)

Reagent Amount to add (for 1 L) Final concentration
Murashige and Skoog (MS) basal salt mixture with vitamins (Phytotechnology Laboratories) 4.3 g 0.43%
Sucrose 30 g 3%
Benzylaminopurine (BAP) 1 mg 0.1%
Indole-3-acetic acid (IAA) 0.1 mg 0.01%
Agar 7.5 g 0.75%
Adjust pH of medium to 5.8 using 1 M NaOH.
Dissolve BAP and IAA in a few drops of 1 M NaOH before adding H2O to make a working solution of 1 mg/mL.
Autoclave and cool to 50°C.
Add filter-sterilized (0.2-μm) antibiotics:
300 mg/L carbenicillin (to kill Agrobacterium LBA4404)
40 mg/L kanamycin (to select for positive transformants)
Pour ~20 mL of medium per dish into Petri dishes and store at 4°C. Medium is good for 1 mo.
