Recipe

XT buffer (pH 9.0)

Reagent Amount to add (for 500 mL) Final concentration
Sodium borate decahydrate (borax) 38.14 g 0.2 M
EDTA 5.71 g 0.5%
SDS 5 mL 1%
Deoxycholate, sodium salt 5 g 1%
H2O to 500 mL
All reagents should be RNase-free. Adjust to pH 9.0 using 1 M NaOH.
Autoclave and store at 37°C to avoid precipitation. XT buffer is good for several years.
