Formaldehyde (5%)
Phosphate-buffered saline (PBS) (10X)
METHOD

1. Add 0.25 g of paraformaldehyde to 4.2 mL of H2O.

2. Add 2 μL of 4 N NaOH.

3. Heat at 65°C until dissolved.

4. Add 0.5 mL of 10X PBS; bring volume to 5 mL with H2O.

5. Filter and store at 4°C for no more than 1 wk.
For C. elegans fixation, concentrations of between 1% and 5% formaldehyde are commonly used; adjust concentration for ideal staining.
