Formaldehyde (5%)

NaOH (4 N)

Paraformaldehyde

Phosphate-buffered saline (PBS) (10X)

METHOD

  • 1. Add 0.25 g of paraformaldehyde to 4.2 mL of H2O.

  • 2. Add 2 μL of 4 N NaOH.

  • 3. Heat at 65°C until dissolved.

  • 4. Add 0.5 mL of 10X PBS; bring volume to 5 mL with H2O.

  • 5. Filter and store at 4°C for no more than 1 wk.

For C. elegans fixation, concentrations of between 1% and 5% formaldehyde are commonly used; adjust concentration for ideal staining.
