Sodium succinate dibasic hexahydrate (Sigma-Aldrich S2378) 0.27 g 5.6 mM
Uracil (Sigma-Aldrich U0750) 0.11 g 5.6 mM
Prepare 0.1-M stock solutions of each component except for the sodium succinate dibasic hexahydrate and uracil by dissolving the appropriate amount of compound in 50 mL of ultrapure H2O and filter-sterilizing. (Sodium succinate dibasic hexahydrate and uracil do not dissolve readily in 50 mL.)
To prepare the cocktail, combine 10 mL of each stock solution with 20 mL of ultrapure H2O (total volume = 180 mL). Add the sodium succinate dibasic hexahydrate and uracil as powders. Stir to dissolve, and filter-sterilize.
The cocktail is used to supplement yeast metabolic footprinting (YMF) medium at a ratio of 9 mL cocktail to 41 mL medium, such that each component of the cocktail is at a final concentration of 1 mM.
