Transfection volume = total transfection volume = volume of vector preparation plus volume of medium covering the cells. As a rule of thumb, nonviral vectors should be prepared in a manner such that the final nucleic acid concentration in the vector preparation does not exceed 100 μg/mL, and preferably does not exceed 50 μg/mL.
