Apple juice-agar plates
0.6 g methyl 4-hydroxybenzoate (Sigma H5501)
10.0 g sucrose
250 mL apple juice
9.0 g Bacto agar
300 mL H2O
Prepare the apple juice solution by adding the first two ingredients to the apple juice. Boil and then let cool.
Prepare the agar solution by adding the Bacto agar to the H2O. Boil.
Mix the apple juice and agar solutions, cool to 65ºC, and pour into the lids of 35-mm Petri plates. (Save the Petri plate bottoms for embryo collection.) This recipe makes 275 plates.
