The desired Ca2+ concentration can be achieved by adding Ca2+/EGTA or Ca2+/BAPTA buffer. Appropriate concentrations of Ca2+ and EGTA/BAPTA can be calculated using widely available calculators (e.g., http://www.stanford.edu/~cpatton/webmaxcE.htm).
