Holding pipettes

Equipment
Capillary tubes, borosilicate glass, 10-cm length, 1-mm OD, 0.5-mm ID (e.g., Clark or equivalent)
Container (clean, airtight)
Ethanol burner
METHOD

1. Using an ethanol burner, gently heat the capillary tubes ~2 cm from one tip.

2. Bend the glass until the desired angle is obtained.
The angle will vary according to the type of micromanipulator used.

3. Carefully flame-polish the capillary opening of the bent tip.
Non-polished capillary openings might damage (cut into) the immobilized blastocyst.

4. Store holding pipettes in a clean airtight container until use.
