10 mM HEPES (pH 7.9)
300 mM KCl
1.5 mM MgCl2
Immediately before use, add:
0.5 mM PMSF (phenylmethylsulfonyl fluoride)
Protease inhibitor cocktail (one tablet; Roche Applied Science 11836170001)
300 μg/mL Flag peptide (Sigma F3290)
Store the Flag peptide in small aliquots at -80°C. Avoid repeated freezing-thawing.
Add the peptide after filtering the buffer using a 0.2-μm bottle-top filter.
