Acetylcholinesterase stain

MATERIALS

Reagents

Acetylthiocholine iodide

CuSO4 (16 mM)

Glycine (80 mM)

Sodium acetate buffer (0.2 M, pH 5.0)

METHOD

Prepare this stain on the day that it will be used.

  • 1. Make an acetylthiocholine iodide solution (Solution A) by adding 0.452 g to 200 mL of H2O.

  • 2. Make Solution B by combining 50 mL of 80 mM glycine, 50 mL of 16 mM CuSO4, and 100 mL of 0.2 M sodium acetate buffer (pH 5.0).

  • 3. Combine Solutions A and B.
