The number of photons needed to give a maximum signal (gray level = 255) on a typical spot-scanning confocal microscope as a function of the gain setting (PMT voltage). On this microscope, typical specimens require a gain setting of 800–900 V, corresponding to ∼180–80 photons/pixel at maximum brightness.
