SC–His Agar Plates

Reagent Quantity Final concentration
Bacto-yeast nitrogen base without amino acidsa 6.7 g 0.67%
Bacto-agar 20 g 2%
Glucose 20 g 2%
Dropout mix (minus histidine) 2 g 0.2%
Prepare 1 L of medium in a 2-L flask.

  • aYeast nitrogen base without amino acids (YNB) is sold either with or without ammonium sulfate. This recipe is for YNB with ammonium sulfate. If the bottle of YNB is lacking ammonium sulfate add 5 g of ammonium sulfate and only 1.7 g of YNB.
