NP-40 Buffer for TAP

Reagent Quantity (for 3 L) Final concentration
Na2HPO4 (FW 142) 2.56 g 6 mM
NaH2PO4·H2O (FW 138) 1.66 g 4 mM
NP-40 (100%) 30 mL 1%
NaCl 26.3 g 150 mM
EDTA (0.5 M) 12 mL 2 mM
NaF 6.3 g 50 mM
Leupeptin 12 mg 4 µg/μL
Na3VO4 (100 mM) 3 mL 0.1 mM
  • Bring to a final volume of 3 L with H2O. Dispense into 500-mL aliquots. Just prior to use, add 10 COMPLETE EDTA-free protease inhibitor tablets (Roche), 1.3 mL of 0.5 M benzamidine, and 5.0 mL of 0.1 M PMSF to 500 mL of NP-40 buffer for TAP.
