CBB Buffer (0.1% and 0.02%)
| Reagent | Quantity (for 100 mL) | Final concentration |
|---|---|---|
| NP-40 (10%) | 1 mL | 0.1% |
| Tris-HCl (1 M, pH 8.0) | 1 mL | 10 mM |
| NaCl (5 M) | 3 mL | 150 mM |
| Magnesium acetate (1 M) | 100 µL | 1 mM |
| Imidazole (1 M) | 100 µL | 1 mM |
| CaCl2 (1 M) | 200 µL | 2 mM |
| β-Mercaptoethanol (β-ME) | ||
| (14.3 M) | 70 µL | 10 mM |
Adjust to a final volume of 100 mL with H2O. Add the β-ME to the CBB buffer just prior to use. The 0.02% CBB buffer is identical to 0.1% CBB buffer, except 200 µL of 10% NP-40 is used.
