Drosophila Adult Hemolymph-Like Saline (AHLS)
| Reagent | Final concentration (mm) |
|---|---|
| CaCl2 | 2 |
| KCl | 5 |
| HEPES | 5 |
| MgCl2 | 8.2 |
| NaCl | 108 |
| NaHCO3 | 4 |
| NaH2PO4 | 1 |
| Sucrose | 10 |
| Trehalose | 5 |
| Adjust the pH to 7.5. Filter. Store at 4°C. Measure the osmolarity of this solution using a freezing-point osmometer. The osmolarity should be 265 mOsm, very close to the value for adult Drosophila hemolymph (Singleton and Woodruff 1994). Use solutions of CaCl2 and MgCl2 instead of powders to avoid precipitation in the saline. | |
