HPLC Buffer A

Reagent Final concentration
KH2PO4 (HPLC grade) 20 mm
Tetrabutylammonium dihydrogen phosphate
(TBAHP) (1 m solution)
 5 mm
To prepare 1 L of buffer A, dissolve 2.722 g of KH2PO4 in a beaker containing ∼800 mL of double-distilled water and add 5 mL TBAHP solution. Adjust the pH to 6 by adding KOH. Transfer the solution into a 1-L volumetric flask. Rinse the beaker repeatedly with water and transfer these rinses into the volumetric flask and adjust the volume to 1 L. Remove any small particles by vacuum filtration using a 0.2-μm PES filter. Sonicate HPLC buffer A for 5 min to release air bubbles. The buffer can be stored for 2–3 d at 4°C. Before use, warm the buffer up to room temperature and sonicate again.
