Buffer D (Dialysis)

Reagent Quantity (for 2 L) Final concentration
Tris–HCl (1 m, pH 7.9)  40 mL  20 mm
Glycerol (100%, v/v)  400 mL 20% (v/v)
KCl (3 m) 66.6 mL  100 mm
EDTA (0.5 m, pH 8.0)  0.8 mL  0.2 mm
Dithiothreitol (DTT; 1 M)   2 mL   1 mm
H2O   to 2 L
Carefully pour the glycerol into the bottom of a 2-L measuring cylinder so that it does not run down the sides. To store buffers for later use, make them without DTT and add fresh DTT to the portions used. Prepare buffers the day before so that they will be well chilled.
