Fluorescence image of a single Cy5 fluorophore. The emitted light (wavelength ∼670 nm) is imaged onto a CCD camera. Although the fluorophore is only nanometers in size, its image appears as a broad spot with an approximately Gaussian intensity profile. The position of the fluorophore can be determined with nanometer precision by fitting the image to find its centroid.
