Schwann Cell Growth Medium

Reagent Amount to add
80% DMEM/20% dH20 18.5 mL
Penicillin/streptomycin (100×) (Gibco 15140-122)  200 µL
Insulin stock (0.5 mg/mL)  200 µL
Sodium pyruvate (100 mm) (Gibco 11360-070)  200 µL
l-Glutamine (200 mm) (Gibco 25030-081)  200 µL
SATO supplement, NB-based (100×)  200 µL
Thyroxine (T3) stock (4 µg/mL)  200 µL
B-27 (Invitrogen) or N21-MAX (R&D Systems)  400 µL

Filter through a 0.22-μm filter and store for up to 1 wk at 4°C. Just before use, add the following:

NAC stock (5 mg/mL) 20 µL
Forskolin stock (4.2 mg/mL) 20 µL
