Recipe

HNEB2 Buffer

Reagent Quantity (for 250 mL) Final concentration
Tris–HCl (1 m, pH 7.5)   2.5 mL  10 mm
NaCl (5 m)     5 mL 100 mm
MgCl2 (1 m)   625 µL 2.5 mm
Triton X-100 (20%)  6.25 mL   0.5%
Phenylmethylsulfonylfluoride (PMSF; Sigma-Aldrich P7626; 0.2 m in isopropanol)   250 µL 0.2 mm
Protease inhibitor cocktail (1000×)   250 µL    1×
H2O to 250 mL
Combine the first four reagents and H2O. Filter with a 0.2-mm filter. Store at 4°C. Add PMSF and protease inhibitor cocktail immediately before use.
