Yeast Sporulation Medium
| Reagent | Final concentration |
|---|---|
| Potassium acetate | 1% (w/v) |
| Raffinose | 0.02% (w/v) |
Prepare sporulation medium (SPM) by combining the ingredients above, adjusting the pH to 7 with 10 n KOH, adding 20 µL of antifoam agent (polypropylene glycol 2000) per liter of solution, and aotoclaving. Supplement SPM with nutrients below according to the auxotrophic requirements of the yeast strain of interest prior to autoclaving.

|---|---|---|
| Adenine | 0.5% | 1.6 mL/L |
| Arginine | 2.0% | 0.2 mL/L |
| Histidine | 2.0% | 0.2 mL/L |
| Isolucine | 1.0% | 0.6 mL/L |
| Leucine | 1.0% | 1.2 mL/L |
| Lysine | 1.5% | 0.4 mL/L |
| Methionine | 2.0% | 0.2 mL/L |
| Threonine | 6.0% | 1.0 mL/L |
| Tryptophan | 1.0% | 0.4 mL/L |
| Tyrosine | 0.25% | 2.0 mL/L |
| Valine | 3.0% | 1.0 mL/L |
| Uracil | 0.2% in 1% Na2CO3 | 1.0 mL/L |
