Fixative Solution for Spheroplasts

| Paraformaldehyde | 3% (w/v) |
| Sucrose | 3.4% (w/v) |
| Heat 25 mL of deionized water to 60°C. Add 0.75 g of paraformaldehyde and 2 drops of 1 n NaOH, and stir with a magnetic stir bar for 5–10 min. Filter sterilize and adjust the pH to 6.5 using pH paper and 1 m HCl. Add 0.85 g of sucrose. Store for no longer than 1 mo at 4°C. | |
