K-Aspartate-Based Pipette Solution

Aspartic acid  130

MgCl2   0.5
MgATP   5
K5Fluo-3 0.05
K-HEPES   10
Adjust the pH of this solution to 7.2 during the titration of aspartic acid with KOH. This solution can be prepared before the experiments and can be stored and kept at –20°C for several weeks. However, it is customary to aliquot and store the solution without the K5Fluo-3, which is added to the pipette solution just before use. (Note that Fluo-3 will buffer contaminating Ca2+ from reagents, so if less Fluo-3 is used, additional buffer [EGTA] should be added.)
