Prokaryotic Elution Buffer

20 mm Tris-HCl (pH 7.5)

300 mm NH₄Cl

10.5 mm MgOAc

0.5 mm EDTA

3 mm β-mercaptoethanol

Prepare in ultrapure (MilliQ) or certified RNase-free water. Filter-sterilize through a 0.22-μm filter. For bacteria other than E. coli, whose exact elution conditions may not be known, buffer with a higher concentration of NH₄Cl may be required, while column elution can be conveniently achieved with a gradient from 0.1 to 1 m NH₄Cl in this buffer.
