Caspase Assay Lysis Buffer

Reagent Final concentration
HEPES (pH 7.5)  50 mm
NaCl 100 mm
EDTA  1 mm
CHAPS  0.1%
Sucrose   10%
DTT   5 mm
Triton X-100  0.5%
Glycerol   4%
Protease inhibitor cocktail (cOmplete, Roche)   1×
After adding the DTT and protease inhibitor cocktail, the lysis buffer can be stored at –20°C in 10-mL aliquots for up to 1 yr.
