Ringer's Solution for Neuromuscular Explants (1×)

Reagent Quantity (for 1 L) Final concentration
CaCl2 (1 m)  2 mL  2 mm
H2O 900 mL
MgCl2 (1 m)  1 mL  1 mm
Ringer's solution (10×) 100 mL   1×
Glucose 3.60 g 20 mm
Prepare CaCl2 and MgCl2 monthly as 1 m stock solutions. On the day of the experiment, mix the reagents in the order listed. Bubble with 95% O2/5% CO2 for 30 min before use. The 1× Ringer’s solution should have a pH of 7.8 without 95% O2/5% CO2 and a pH of 7.3 after 30 min of bubbling with 95% O2/5% CO2.
