Synthetic Complete (SC) Agar Medium for OyCD PCA

Reagents	Final concentration (1×)
Yeast nitrogen base (YNB) without amino acids and without ammonium sulfate	1.74 g/L
Agar (for solid media only; see below)	2%/3% (w/v)
Amino acid mix (10×), without histidine or leucine	1×
Glucose (if required; see below)	2% (w/v)
Raffinose (if required; see below)	2% (w/v)
Galactose (if required; see below)	2% (w/v)
G418 (if required; see below)	200 µg/mL (w/v)
5-Fluorocytosine (5-FC) (if required; see below)	1 mg/mL (w/v)
Use 3% (w/v) agar in OmniTray plates or 2% (w/v) for Petri dishes. Prepare the following formulations: (1) solid medium in Petri plates with glucose; (2) solid medium in Omniplates and liquid medium with glucose and G418; (3) liquid medium with raffinose; (4) and solid medium in Omniplates with raffinose, galactose, and G418, with and without 5-FC. For long term storage, keep at 2°C–8°C.
