Synthetic Complete (SC) and Synthetic Defined (SD) Media for PCAs

Recipe

Reagents	Final concentration (1×)
Yeast nitrogen base without amino acids and with ammonium sulfate	6.7 g/L
Agar (for solid medium only)	2% (w/v)
Amino acid mix (10×), without histidine and leucine (for RLuc and Venus PCA experiments) or without uracil and histidine (for IFP PCA experiments)	1×
Glucose (if required)	2% (w/v)
Raffinose (if required)	2% (w/v)
For SD, omit the amino acids, glucose, and raffinose. For SC, the addition of glucose or raffinose depends on the point in the procedure in which it will be used. Sterilize by autoclaving and store at room temperature.
